This companion to Cells! is copyright (2004) R.Howard Berg (rhberg@danforthcenter.org) and the Donald Danforth Plant Science Center, St. Louis, Missouri, U.S.A. (danforthcenter.org) and is distributed for educational purposes only—we retain the sole rights to distribute and permission is granted to use these images for educational purposes only, and are not to be sold or published without the written permission of the Donald Danforth Plant Science Center. We are grateful to the Major Research Instrumentation Program of the National Science Foundation for funding the acquisition of the Zeiss LSM 510 META NLO multiphoton microscope used in making these images, and to NASA for funding the purchase of the Coherent Mira 900F titanium-sapphire pulsed laser that served as the two-photon excitation source.

These images are digital 3D reconstructions of optical sections made by two-photon excitation of cyan fluorescent protein (CFP) localized in the endoplasmic reticulum (ER-CFP) of these tobacco BY-2 cultured cells. The Imaris module of Bitplane software was used in making the reconstructions. For viewing the color images in 3D (stereo depth perception) you must use red/cyan viewing glasses (red view through the left eye, cyan view through the right eye).

For playback of the movies, Quicktime for PC or Mac is free and available from the Apple website—we recommend using this software for the movies.

The images shown were produced using instrumentation in the Integrated Microscopy Facility
at the Danforth Center and illustrate a variety of cellular characteristics and structures.
