Propagation of Cassava (Manihot esculenta Crantz) by the Temporal Immersion System

        For the first time, we report use of the Temporal Immersion System (TIS) in the massive propagation cassava by organogenesis. This research was carried out in the Biotechnology Lab. of the Research Institute in Tropical Root and Tubers Crop (INIVIT) in order to establish a methodology for the cassava micropropagation in Temporal Immersion System. Experiments have been made in order to increase the multiplication coefficient where conventional micropropagation method, the stationary medium of culture liquid and in agitation, and the TIS were compared. Different variants of the medium MS (1962) were also studied, as well as different times of immersion, volume of medium for explants and density for the material to a same immersion frequency. 'CMC-40' clone from the Institute germplasm bank was used. The evaluations were carried out to the 30 and 40 days of cultivation according to the experiments. The fresh weight, active leaf numbers, length and the cutting numbers of the explants, as well as the multiplication coefficient were evaluated. The obtained results permitted to establish a efficient methodology for the cassava micropropagation based on the use of the TIS to increase in a significant way the multiplication coefficients and the production of in vitro plants with improved quality for acclimatization to field conditions.
